You'd be crazy to come to Vietnam and not do the Mekong Delta tour. It's best to do it over 2 or 3 days with a possible stop over in Cantho. The bus boat combination worked well for me although I expect doing all the trip by boat would be particularly rewarding. You can also do a homestay in the Mekong Delta in a traditional house. You could probably spend a month in the Mekong and still be longing for more.
